本辣根过氧化物酶标记Streptavidin(HRP-labeled Streptavidin)也称HRP-Streptavidin、Streptavidin-HRP或辣根过氧化物酶标记链霉亲和素,为进口分装,可以用于生物素(Biotin)标记的抗体、核酸、蛋白或其它生物素标记分子的检测。具体用途包括Western、ELISA、免疫组化或免疫细胞化学、原位杂交、Southern、Northern、EMSA等。
本HRP-Streptavidin用高纯度的Streptavidin和超纯的辣根过氧化物酶交联而成,确保产生最低的本底和最高的灵敏度。本产品的浓度为1mg/ml。
Streptavidin的分子量为66kD,可以高度特异性地和生物素(Biotin)结合。Streptavidin和生物素的亲和常数为 Kd=10-15M。Streptavidin是一个4聚体蛋白,可以同时结合4个生物素分子。Streptavidin的中文名为链霉亲和素,从Streptomyces adidinii中纯化获得,和鸡蛋清来源的Avidin(亲和素)在空间结构以及与生物素的亲和力方面具有高度的相似性。和Avidin不同的是,Streptavidin是一种非糖基化蛋白,并且基本不带电荷。Avidin的pI=~10.5,在中性pH条件下呈碱性。由于Streptavidin和Avidin相比在中性条件下不带电荷,因此Streptavidin的非特异性结合比Avidin低很多,这样检测时的非特异性背景就低很多。因此目前在生物素检测时通常使用Streptavidin替代Avidin。
辣根过氧化物酶即horseradish peroxidase(HRP),可以在Western、EMSA、Southern或Northern等检测时催化ECL类试剂(如ECL、BeyoECL等)产生化学发光,可以在ELISA时催化TMB产生蓝色,也可以在免疫组化、免疫细胞化学或Western blot检测时催化DAB产生棕色沉淀。
本辣根过氧化物酶标记Streptavidin用于各种常规用途的推荐稀释比例参考下表,实际实验操作过程中需根据具体情况适当调节辣根过氧化物酶标记Streptavidin的稀释比例。对于Western,推荐的稀释范围为1:2000-10000。
